How Does HT Mechanoporation Work?

Our Galaxy systems work by boosting cells as they transit through the membrane onto plates with pre-plated cargo where they then take up the material

How It Works

This works because after boosting the pores in the cell membrane remain open for approximately 30 seconds. During this brief window, cells are dispensed directly onto plates with pre-plated cargo, enabling efficient uptake of materials.

Key Advantages

  • Pre-loaded plates: prepare cargo libraries in standard plates ahead of time
  • On-demand processing: Add cells and process when ready – no wait time for cargo prep
  • Multi-cargo flexibility: Different payloads per well for screening

Critical Timing Window

Cell membrane pores remain open for 5–30 seconds post-boosting, allowing efficient cargo uptake when cells are dispensed onto pre-plated material. This brief window enables high-throughput processing while maintaining cell viability and function.

Platform Validation Data

Primary Cell Crispr Screening

Demonstrated proof-of-concept for CRISPR screening in primary cells using the Galaxy-i with Certus Flex. This workflow achieved successful B2M RNP delivery to primary T cells in a high-throughput system—enabling functional genomics applications in clinically relevant cell types.

Consistent Delivery Across Plates

Validated consistent delivery performance in HeLa cells across five 96-well plates using diverse cargo types: 3kDa dextran, labeled oligo, and IgG antibody—demonstrating platform reliability across molecular sizes and formats.

Broad Cell Type Compatibility

Demonstrated successful delivery across multiple cell types, including HEK293 cells (oligo & antibody) and human peripheral blood mononuclear cells (PBMCs shown here)—critical for drug discovery programs that require validation across diverse cell models.

Complex Cargo Delivery

Achieved high GFP mRNA transfection efficiency in HeLa cells while preserving cell viability—demonstrating the platform's capability for delivering large, complex cargos essential for gene editing, regulatory RNA studies, and functional genomics screening.
